بروتين NLRP3 نشط في كل من مخ البشر والفئران





تمكن باحثون بالمركز الألماني لأمراض التحلل العصبي وجامعة بون من اكتشاف أن بروتين NLRP3 يمثل عامل أساسي في تطور مرض الألزهايم, ويلعب هذا الفيتامين دوراً هاماً في الجهاز المناعي, فعند تفعيله يتم تحفيز الخلايا المناعية ويتم أطلاق المواد التي تسبب الالتهابات.

وقد ركزت دراسات متعددة وضعت خلال الأعوام القليلة الماضية علي كيف ترفع الاستجابة المناعية للجسم من مخاطر الإصابة بالألزهايمر وكيف أن جينات بعينها تتدخل في تلك العملية.

كما وجدت دراسة أخري أن التحور الجيني في جين TREM2 الذي يحفز استجابات الجهاز المناعي يرتبط أيضاً بمرض الألزهايمر.

وبمجرد أن يتم إثارة جهاز في مرض الألزهايمر الإحساس يتسبب ذلك في موت الخلايا العصبية الأمر الذي ينتج عنه فقدان وظيفة المخ والقدرة المعرفية.

ويحدث مرض الألزهايمر عندما تبدأ البقع النشوية في التشكل في المخ وتعمل هذه البقع علي تفعيل بروتين NLRP3. في هذه الدراسة نظر الباحثون في تأثير هذه الزيادة في نشاط البروتين علي المخ.

ويعتقد الباحثون أنه بكبح رد الفعل الإلتهابي لهذا البروتين سوف يكون هناك تناقص للبقع المدمرة للمخ, وقد قاموا بإزالة الجينات التي تثير النشاط الإلتهابي لبروتين NLRP3في مجموعة من فئران المعمل تعاني الإضطرابات المرتبطة بالألزهايمر.

وبعد إزالة الجين لم تعد الفئران قادرة علي إنتاج مركب البروتين, وهذا ينتج عنه بعض الأعراض الخفيفة فقط من المرض.

ويمكن أن تؤدي هذه الأخبار الواعدة للوصول إلي عقاقير دوائية قادرة علي استهداف هذا المكون البروتيني وتثبيط نشاطه كما يري الباحث.

وصرح البروفسور ميخائيل هنيكي أن فريقه يعمل بالفعل علي الوصول لطرق لكبح هذا البروتين لقد تعثرنا أمام عامل هام في تطور المرض, بالوصول لتلك النتائج يبدو أن هناك إمكانية لكبح نشاط الإلتهابات التي يسببها بروتين NLRP3, في الوقت الحالي هناك اختيارات متعددة يمكن أن نتبعها للتعامل مع المرض أثناء مساره.

تشير نتائج هذه الدراسة لإمكانية جديدة إلا أننا لازلنا في مرحلة الدراسة.
